    PATHOMORPHOLOGICAL CHANGES IN LAYERS OF RETINA FOR SIX WEEKS OF OPIOID EXPOSURE EXPERIMENT

    Annotation The objective set in the given study wasto examine pathomorphological manifestations of angio-and neuroretinopathy as a result of chronic exposure to therapeutic doses of opioid analgesics. Sexually mature outbred male rats in the amount of 15 animals weighing 190 grams, age 4.5 months served as study materials. Animals were injected intramuscularly with nalbuphine daily once a day in the same period of time (10-11 am) for 42 days. The initial dose of nalbuphine during the first 2 weeks was 0.212 mg/kg, the next 2 weeks (II - IV week) 0.225 mg/kg, and during IV- VI weeks - 0.252 mg/kg. Thus, chronic opioid effect was obtained [20]. Animals were divided into 2 groups. Group 1 animals received nalbuphine for 42 days, followed by sampling of study material (end of 6th week of experimental opioid effect); Group 2 of controls that was injected with normal saline intravenously for 42 days in the same period of time (10 - 11 am). All animals were in vivarium and conditions that dealt with issues of maintenance, care, labelling and all other manipulations were carried out in compliance with the provisions of “European Convention for the Protection of vertebrate animals used for experimental and other scientific purposes” [Strasbourg, 1985], “General ethical principles of animal experimentation” adopted by the first National Congress on Bioethics [Kyiv, 2001], the Law of Ukraine No3447 – IV “On prevention of cruelty to animals”. Bioethics Committee of Danylo HalytskyLviv National Medical University determined that the given study met the ethical requirements according to MOH of Ukraine No 231 of 01.11.2000 (Record No 10 of 26.12.2011), (record No 2 of 20.02.2012). Before sampling of material vital animals were narcotized by intraperitoneal administration of sodium thiopental (calculated as 25 mg/kg). The rats’ eyeballs were used as the material for microstructural research obtained by postvital enucleation with further actions to preserve topographic relationship of eye membranes. Histological sections 5 - 7 microns thick were produced. Histologic specimens were prepared by the conventional method dyed with hematoxylin, eosin and azane by Heidenhain. Microscopic examination and photographic recording of specimens were carried out by means of a microscope MBI-1 and digital camera Nikon D3100. Results of the study in the future will allow us to form pathomorphological database that can be used to conduct comparative characteristics. Concerning the processes of the increase of pathological changes in the layers of retina during short-term and long-term effects of small doses of opioids, the above-mentioned will enable further development of methods for optimal correction of angio- and neuroretinopathy caused by opioid effects.
